There were different sites:

Berkeley Labs where Lawrence and his group were using cyclotrons for enrichment.

Handford where Pu was being manufactured via neutron activation of U-238.

Oak Ridge using gaseous diffusion for enrichment.

I think MIT's Rad Lab was part of it.
