DNA Ligation

    Is there anyway by which one can selectively prevent 3' end or 5' end of the DNA from ligation?

    End capping DNA, one way is by dephosphorylating the phosphate, with CIP (calf intenstestine phophatase). There are other methods available: search for "end capping dna".
