If I wish to calculate the energy of a collision between two bodies do I just add the velocities of each body and use E = mc^2?

You should provide more detail. What do you mean by "energy of a collision"? I understand that you want to discuss the collision of two bodies, so please start from there, and explain what you want to calculate.
