SUMMARY

This discussion addresses the determination of DNA weight in samples for genomic extraction, specifically using HeLa and human fibroblast cell lines. The protocol for genomic DNA extraction requires DNA solutions to be less than or equal to 1 mg/ml. Participants recommend using DNA quantitation assay kits from Life Technologies or Sigma-Aldrich to accurately measure DNA concentration. If the sample exceeds the linear range of the assay, users should dilute the sample accordingly and track the dilution factor to ascertain the original concentration.

PREREQUISITES
  • Understanding of genomic DNA extraction protocols
  • Familiarity with DNA quantitation assay kits
  • Knowledge of sample dilution techniques
  • Basic laboratory skills for handling cell cultures
NEXT STEPS
  • Research the Life Technologies DNA Quantitation Assay Kit for specific usage instructions
  • Explore the Sigma-Aldrich DNA Quantitation Assay Kit for alternative options
  • Learn about proper dilution techniques for accurate concentration measurement
  • Investigate common pitfalls in genomic DNA extraction protocols
USEFUL FOR

Researchers, molecular biologists, and laboratory technicians involved in genomic DNA extraction and quantification will benefit from this discussion.

This is my first time isolating the whole genomic DNA from cells. I have two cell line HeLa and Human fibroblasts (almost fully confluent T-25 flasks). The problem is that the protocol which is to be followed for genomic dna extraction is only suitable for DNA solutions less than or equal to 1mg/ml (according to the instructions). How do I determine the weight of DNA that I have beforehand? I'm confused on that point. (Note I can't count cells either).
