why reflection coefficient R=1 in step potential though there happens transmission also as a negative exponential when E<V0

There is no transmission when E<V0 for a step potential ... that would mean the particle was traveling inside the classically forbidden region ...
 
The coefficients R and T are determined by the probability of an incident particle traveling to - or + infiity.
