Hi

I am not sure whether the object plane overclaps focal plane in a infinity-corrected optical microscope.

I mean when the miscroscope is well focused. Is it the object that in the focal plane can be best imaged?
 
If I understand you correctly, yes. When the microscope is properly aligned, the specimen plane is located at the front focal plane of an infinity-corrected objective.
