Do power plant generators regulate the output voltage the same way a car alternator regulates its output voltage. Or do power plant regulators control the output by regulating the rotation speed of the generator.

yes, exactly same principle - vary the field to control voltage .
Very small plants like windmills may use another method, so above is a generalization but a pretty true one and i think in the spirit of your question.

Or ... by regulating the rotation speed of the generator

Think about it for a minute - if the speed changed so would frequency.

I am trying to understand how transferring electric from the powerplant to my house is more effective using high voltage. The suggested explanation that the current is equal to the power supply divided by the voltage, and hence higher voltage leads to lower current and as a result to a lower power loss on the conductives is very confusing me. I know that the current is determined by the voltage and the resistance, and not by a power capability - which defines a limit to the allowable...
