SUMMARY

Detergent lysis is an effective method for isolating proteins from biological samples, particularly when harsh chemicals are restricted. In this discussion, Tom seeks to analyze protein contents in various fish species but faces challenges in lysing samples without using phenol-chloroform extraction. He plans to quantify protein concentration using the Biuret test or Ninhydrin in conjunction with a photometer. The discussion emphasizes the importance of selecting appropriate lysis methods that comply with safety regulations.

PREREQUISITES
  • Understanding of protein extraction techniques
  • Familiarity with Biuret and Ninhydrin tests
  • Knowledge of photometric analysis
  • Basic principles of molecular biology
NEXT STEPS
  • Research detergent lysis protocols for protein extraction
  • Learn about alternative lysis methods suitable for sensitive samples
  • Explore the Biuret test and its application in protein quantification
  • Investigate the use of Ninhydrin for amino acid detection
USEFUL FOR

Students in biology or biochemistry, researchers focusing on protein analysis, and anyone interested in safe methods for protein extraction from biological samples.

TL;DR Summary: I have to do a project for my school, and I want to know how it works, because I just can't get any useful information of the internat alone.

I have a project for school, I want to analyse and compare the protein contents of some different species of fish. The problem I'm stuck with, is just separating the protein from nucleic acids and other organic matter. I want to get the concentration by either using the Biuret test, or using Ninhydrin so that I can use a photometer to get that concentration. But still I am either stuck with lysing my sample, and getting that protein, especially with the regulations we have at my school, I cannot use hard chemicals e.g. I can't do a phenol-Chloroform extractionl.
