Why Does Fluorescence Decrease During the Lag Phase in 3T3 Cell Growth Curves?

AI Thread Summary
The discussion centers on an observed decrease in fluorescence values during the lag phase of a cell growth curve using 3T3 cells. The individual conducting the experiment is puzzled by this phenomenon, as it contradicts typical expectations and lacks clear explanations in existing literature. They speculate that cell death might be occurring at a higher rate than replication during the attachment phase, but this seems unlikely given the timing of the decrease. Participants in the discussion suggest conducting a statistical analysis to determine if the decrease is significant or merely a result of normal variance or experimental noise. They also inquire about the availability of raw data for further evaluation.

Hi All,

Having plotted a cell growth curve (fluorescence vs days), I have found that during the lag phase, the fluorescence values decrease. I haven't come across this previously during lectures and can't seem to find any examples online to help explain it. My only thought was that cell death occurred at a higher rate than replication whilst the cells were attaching to the culture flask but the decrease occurs between 1 and 3 days so this doesn't seem like a possible explanation. As a side note, 3T3 cells were being used during the experiment.

I'd appreciate any advice on this or resources that may be useful.

Many Thanks

Have you run statistical analysis on the fluorescence values? Is it statistically significant decrease? Can you provide the raw data? It could just be normal variance or noise that you're interpreting as a decrease.

I am reading Nicholas Wade's book A Troublesome Inheritance. Please let's not make this thread a critique about the merits or demerits of the book. This thread is my attempt to understanding the evidence that Natural Selection in the human genome was recent and regional. On Page 103 of A Troublesome Inheritance, Wade writes the following: "The regional nature of selection was first made evident in a genomewide scan undertaken by Jonathan Pritchard, a population geneticist at the...
